Trimix FAQs

The Plan
How many units of Trimix should I take?

Trimix dosage can vary widely from person to person and there are several variables that help your doctor to determine a good starting point. Most patients start by using around 10 to 15 units of the ED injectable they were prescribed and then increase the dosage by small increments each usage until a hard sustained erection is achieved.

How often can I use Trimix injections?

Most patients can inject Trimix up to three times per week, with 24 hours between each injection.

Does Trimix work better than Viagra?

Trimix is more effective than Viagra based on the percentage of men who report their ED is improved. The success rate for Trimix is 80% to 95%. Viagra is around 60% to 70% effective for men with moderate ED.

More men use Viagra than Trimix because taking a pill is easier than doing a penile self-injection. However, men using Trimix report that the injection procedure is nearly painless, and the results are fast and consistent.

Many men, especially those taking medications for heart disease, cannot take Viagra or other medications in that class. Trimix is a great option for these men and men who want an ED treatment option that is not affected by diet or alcohol consumption.

Are injections better than pills for ED?

The American Urological Association recommends a process of shared decision-making when deciding which treatment option to use for ED. Men should be offered a full range of treatment options. Some men have medical conditions or are taking medications that preclude them from taking ED pills. For these men, ED injections are a highly effective option.

Do Trimix injections lose effectiveness?

Trimix may not work for all men, including men with the following conditions:

  • Penile scarring
  • Peyronie’s disease
  • Poorly controlled diabetes
  • Dialysis-dependent kidney disease
  • Severe venous leak syndrome

Talk to your healthcare provider if Trimix does not work for you. You may need a dosage change. They may also need to review your injection technique to ensure the medication is being delivered into the corpora cavernosa. Also, check the expiration date on your Trimix and ensure that you are storing it correctly.

What is the difference between Trimix and Caverject?

Trimix is three medications that work synergistically to increase blood flow into the penis and produce an erection. The three medications in Trimix are:

  • Prostaglandin
  • Papaverine
  • Phentolamine

Caverject only contains prostaglandin.

What happens if you inject Trimix too often?

Trimix should always be injected following your healthcare provider’s instructions. If Trimix is injected too frequently or at too high a dose, it could cause priapism.

Priapism is an erection that lasts longer than four hours. A prolonged erection allows blood to sit in the penis for too long. Fresh blood is needed to replenish oxygen and nutrients and remove waste products. Untreated priapism can damage the penis.

How often can I inject Trimix?

It is important to follow the instructions provided by your healthcare provider and pharmacist.

In general:

  • Trimix should never be injected more than once in a 24-hour period.
  • Trimix should not be injected on more than two consecutive days.
  • Trimix should not be injected more than three times in one week.

This process includes an online medical consultation, prescription approval, the medication getting filled at the pharmacy, and the medication being sent to your home.

Trimix is a Combination of Three Medications That Increase Blood Flow Into the Penis

Trimix is an injectable medication used to treat erectile dysfunction. It is a formulation of three drugs: phentolamine, papaverine, and alprostadil (prostaglandin E1), making it an excellent ED treatment alternative for men who cannot or do not want to take phosphodiesterase inhibitors, have severe side effects from oral ED medications, or have not had successful results when taking oral ED medications.

The three medications in Trimix work synergistically to increase blood flow to the penis. Based on clinical studies, the success rate for Trimix when treating ED approaches 90%.

The American Urological Association reported that 53.7% to 100% of men (depending on population) achieved a satisfactory erection with Trimix, and 46.3% to 98.8% of men were happy with their choice to use intracavernosal injections as a treatment option.

Phentolamine

The alpha-adrenergic antagonist phentolamine relaxes the smooth muscle lining the corpora cavernosa. It is commonly used in conjunction with papaverine to treat ED. Clinical studies show that phentolamine, when combined with papaverine, dilates blood vessels entering the penis, resulting in an erection in 20 to 30 minutes.

Papaverine

Papaverine, one of the first injectable medications used to treat ED, is a vasodilator, which means that it dilates blood vessels and increases blood flow into the penis. Papaverine is no longer used to treat ED on its own. It is used in conjunction with phentolamine and prostaglandin to improve treatment outcomes while minimizing side effects.

Prostaglandin E1

Prostaglandin E1 relaxes smooth muscle surrounding blood vessels in the penis and the corpora cavernosa in the penis, allowing them to fill with blood. Blood becomes entrapped in the penis, which leads to an erection. Prostaglandin can be administered by intracavernosal injection or as pellets inserted into the urethra.

Bimix and Quadmix

Although Trimix is a frequently prescribed injectable to treat ED and it is highly effective, it is not the only option.

Bimix: Bimix is a combination of phentolamine and papaverine. It is an excellent choice for men with sensitivity to prostaglandin E1 or who have health conditions that contraindicate its use.

Quadmix: Quadmix combines phentolamine, papaverine, atropine, and alprostadil (prostaglandin E1). Trimix is up to 95% effective in treating ED. If you are in the 5% of men, Quadmix may be a great option.

How Does Trimix Work?

Trimix Increases Blood Flow to the Penis

The three ingredients in Trimix – Alprostadil, Papaverine, and Phentolamine – work in combination to increase blood flow to the penis, which causes a hard erection.

Papaverine Increases Blood Flow into the Penis

Papaverine is an alkaloid that stimulates the production of cyclic adenosine monophosphate (cAMP), a molecule that relaxes the smooth muscle in the walls of blood vessels and lining the corpora cavernosa. Its mechanism is not completely understood because it appears to act on several pathways.

When papaverine is injected into the intracavernosal space of the penis, it increases blood flow, producing an erection. Papaverine also relaxes smooth muscle lining the corpora cavernosa in the penis, allowing them to fill more completely.

Prostaglandin E1 (Alprostadil) Increases Blood Flow into the Penis

Prostaglandin E1 (alprostadil) is a synthetic form of prostaglandin, a potent vasodilator. Prostaglandin E1 relaxes the smooth muscle lining the corpora cavernosa in the penis by activating prostaglandin receptors and subsequently increasing cyclic monophosphate levels in the smooth muscle. This causes a decrease in calcium levels in the smooth muscles, allowing them to relax and fill with blood.

Prostaglandin E1 also relaxes the smooth muscle lining blood vessels that enter the penis, allowing for greater blood flow into the penis.

Phentolamine Increases Blood Flow into the Penis

Phentolamine is an alpha-adrenoceptor antagonist, which means that it blocks chemicals such as epinephrine and norepinephrine from binding to alpha-adrenergic receptors. Competitively binding to alpha-adrenergic receptors, phentolamine causes smooth muscle relaxation and blood vessel dilation causing penile erection. When blood vessels are widened, blood pressure decreases, and blood flow into the penis increases.
